This study reveals that hypoxia in centrally located EGFR-mutant lung adenocarcinomas drives an alveolar lineage imbalance via the ZAKα-MAPK-c-Fos pathway, promoting early tumorigenesis. Therapeutic oxygenation can restore lineage balance and limit aggressive progression.
